I'm presenting my progress to the lab
I'm trying to induce 2 HP1 gene mutations into the fly genome through a gene-editing technique called Homology Directed Repair that requires a conserved sequence of the actual wild-type (Normal) gene so that we hope our mutation is repaired into the genome through recombination. This is after an endonucleade known as Cas9 cuts the double stranded DNA of the HP1 gene to allow for the repair repair to occur.
And I'm currently modifying and replicating a plasmid DNA (bacterial) from a strain of E. Coli to try and create our mutant DNA that contains abconserved sequence but the mutation.
I'm also then doing just typical fly experiments regarding the HP1C protein specifically. I'm taking flies with either a completely absent HP1C protein or ineffective but present HP1C protein and seeing how that affects fly Fertility and Hatch Rate to determine the organisms fitness in development.
And then I'm drafting a grant for RNA-sequencing for the fly work above.
